Premenstrual syndrome (PMS) involves physical and emotional symptoms in the luteal phase of your menstrual cycle, caused by fluctuating estrogen and progesterone levels affecting neurotransmitter systems. After ovulation, progesterone rises then falls sharply before menstruation. This affects your GABA receptors (reducing calming signals), serotonin production (affecting mood), and inflammatory signaling. Your brain's emotional regulation centers—amygdala, hippocampus, prefrontal cortex—have estrogen and progesterone receptors that modulate their activity.
